Type of Immunization Offered
mRNA

Encapsulated mRNA immunization represents the latest advancement in immunization technology and is offered by Immune Biosolutions.
Rather than administering a purified protein, this approach delivers messenger RNA (mRNA) encoding the target antigen. Once taken up by the animal’s cells, the mRNA is translated into the native protein, allowing the immune system to recognize the antigen as it is naturally produced. This strategy can promote robust immune responses while simplifying the development of challenging or difficult-to-produce targets.
It is a very attractive method for large, complex and membrane proteins for which commercial options are unavailable, and when other immunization methods don’t guarantee proper folding of the protein.
DNA

DNA immunization is performed by plasmid injection followed by electroporation to facilitate cellular uptake and antigen expression. Although mRNA immunization has become the platform of choice for many antibody discovery campaigns, DNA immunization remains well suited for applications requiring prolonged antigen expression, enhanced storage stability, or compatibility with established DNA-based workflows.
Proteins

Protein immunization has been the standard for decades. It is a well known and optimized method typically yielding high titers and diverse libraries.
For average targets, when properly folded proteins are available, it is an attractive and safe option that will typically yield excellent results.
Virus-Like Particles

VLPs do not contain a viral genome, are non-infectious and simple to manufacture. They are therefore a very attractive option for viral proteins for safety and regulatory reasons.
VLPs can be enveloped and non-enveloped, and the choice between the two depends on the protein used for immunization. For example, complex viral glycoproteins will benefit the most from the use of enveloped VLPs, while non-enveloped VLPs will perform well for soluble proteins and peptides.
Enveloped VLPs produced in human cells acquire the membrane of the host cell and therefore, they can display human targets with native post-translational modifications and assembly on their surface. Enveloped VLPs can replace whole cells for complex or multipass membrane proteins.
Macrocyclic Peptides

Macrocyclic peptide immunization bridges the gap between linear peptides and full-length proteins by presenting structurally constrained epitopes that more closely resemble their native conformation.
These peptides are well suited for antibody discovery campaigns targeting loop- or turn-dependent epitopes that are poorly represented by linear peptides, particularly when production of the full-length antigen is beyond current technical capabilities or is otherwise impractical.
Linear Peptides

Linear peptides provide a simple and cost-effective solution for targeting continuous epitopes. Their ease of synthesis enables the generation of large, highly diverse peptide libraries for antibody discovery and screening.
Species
Our team brings years of experience in mononuclear cell isolation from whole blood, bone marrow and spleen for the generation of high-quality immune libraries. Our experienced scientists and standardized procedures ensure the consistent production of high-quality libraries while preserving the natural diversity of the immune repertoire in multiple species.
Chickens
Chicken-derived antibody discovery campaigns use bone marrow and spleen as the starting material for immune library generation.
Antibody Discovery
RNA or cDNA
RNA extraction followed by PCR-generated cDNA is part of our standard pipeline and can be shipped directly to our clients.
Splenocytes and BMMCs
We also routinely isolate mononuclear cells from the spleen or bone marrow, which can then be frozen for later use.

Alpacas
Alpaca-derived antibody discovery campaigns use whole blood as the starting material for immune library generation. The volume and timing of blood collection can be tailored to the specific requirements of each project.

PBMCs
We also routinely isolate PBMC fraction from whole-blood, which can then be frozen for later use.
